KERATINOCYTES

Epidermis was the first tissue to be cultured in vitro and transferred back to the patient as an autograft. Since then, the technique has dramatically improved the survival of patients with large burns and is used in most large burn centres to day. At the Karolinska University Hospital, we were among the first in the world to use cultured epidermal autografts and are today using this technique on a routine basis.

Even though this method has had a striking effect on the survival of patients with large burns, the quality of the resulting skin has been disappointing because of the lack of a dermal component and thereby an inadequate strength and durability. Because of this, we have recently developed a method for regeneration of the dermal part of the skin by using collagen templates. Dermal regeneration are today used together with cultured, autologous keratinocytes either as grafts or as single cell suspensions.

The increased quality resulting from this improvement of the methods has resulted in the possibility to use regeneration of skin in other groups of patients than large burns such as minor burns, scar revisions, chronic wounds and skin cancer and other skin diseases that needs excision.
